Test Principale: This assay employs a two-site sandwich ELISA to quantitate IL18BP in samples. An antibody specific for IL18BP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyIL18BP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IL18BP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL18BP bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview: IL18BP is an inhibitor of the proinflammatory cytokine IL18. This protein binds to IL18, prevents the binding of IL18 to its receptor, and thus inhibits IL18-induced IFN-gamma production. This protein is constitutively expressed and secreted in mononuclear cells. The expression of this protein can be enhanced by IFN-gamma. An elevated level of this protein is detected in the intestinal tissues of patients with Crohn's disease. Three transcript variants encoding the same protein have been found for this gene.Sequence analysis revealed that IL18BP encodes a 192-amino acid soluble protein with a 28-amino acid signal sequence. By analysis of a genomic clone, the authors identified 3 splice variants, which they termed IL18BPa, IL18BPb, and IL18BPc.
Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
