Test Principale: This assay employs a two-site sandwich ELISA to quantitate PADI6 in samples. An antibody specific for PADI6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPADI6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PADI6 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PADI6 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview: Peptidylarginine deiminases (PADs) convert arginine residues in proteins into citrullines. Peptidylarginine deiminases (PADs, EC 3.5.3.15), including PADI6, make up a family of posttranslational protein modification enzymes that convert arginine residues to citrulline residues in the presence of calcium ions. PADI6, The deduced 694-amino acid protein has a calculated molecular mass of 77.7 kD. RT-PCR detected expression in adult human tissues, including ovary, peripheral blood leukocytes, and testis, with weaker expression in small intestine, spleen, lung, liver, and skeletal muscle. No expression was detected in other tissues examined. The order of the genes is p-telomere--PADI2--PADI1--PADI3--PADI4--PADI6. Mouse Padi6 maps to a syntenic Pad cluster on chromosome 4E1.
