Test Principale: This assay employs a two-site sandwich ELISA to quantitate RUNX2 in samples. An antibody specific for RUNX2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyRUNX2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for RUNX2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of RUNX2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview: Cbfa1/Runx2 is a key transcription factor associated with osteoblast differentiation.This protein is a member of the RUNX family of transcription factors and has a Runt DNA-binding domain. It is essential for osteoblastic differentiation and skeletal morphogenesis and acts as a scaffold for nucleic acids and regulatory factors involved in skeletal gene expression. The protein can bind DNA both as a monomer or, with more affinity, as a subunit of a heterodimeric complex. Transcript variants of the gene that encode different protein isoforms result from the use of alternate promoters as well as alternate splicing.The RUNX2 transcription factor is the master regulator of osteoblast differentiation is required for chondrocyte hypertrophy .
