Test Principale: This assay employs a two-site sandwich ELISA to quantitate TNRC6B in samples. An antibody specific for TNRC6B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyTNRC6B present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNRC6B is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNRC6B bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview: The deduced 1, 203-amino acid protein contains an RNA recognition motif. RT-PCR ELISA detected moderate expression in all tissues and specific brain regions examined, with highest expression in cerebellum. Schneider et al. (2006) stated that the 1, 727-amino acid TNRC6B protein contains a glycine/tryptophan (GW) -rich N-terminal domain, a central glutamine-rich region, and a C-terminal RNA recognition motif. TNRC6B copurified with a cytoplasmic HeLa cell protein complex containing AGO2 (EIF2C2), DICER, and MOV10. Epitope-tagged TNRC6B coimmunoprecipitated with tagged AGO1 (EIF2C1) and tagged AGO2. TNRC6B was required to mediate microRNA-guided mRNA cleavage in HeLa cell culture.
Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
