Test Principale: This assay employs a two-site sandwich ELISA to quantitate POMC in samples. An antibody specific for POMC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPOMC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for POMC is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of POMC bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview: Pro-opiomelanocortin (POMC) is a precursor polypeptide with 241 amino acid residues. This gene encodes a polypeptide hormone precursor that undergoes extensive, tissue-specific, post-translational processing via cleavage by subtilisin-like enzymes known as prohormone convertases. There are eight potential cleavage sites within the polypeptide precursor and, depending on tissue type and the available convertases, processing may yield as many as ten biologically active peptides involved in diverse cellular functions. The encoded protein is synthesized mainly in corticotroph cells of the anterior pituitary where four cleavage sites are used; adrenocorticotrophin, essential for normal steroidogenesis and the maintenance of normal adrenal weight, and lipotropin beta are the major end products.
Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
