Poliovirus receptor-related immunoglobulin domain-containing (PVRIG), also known as CD112 receptor (CD112R), is a 34 kDa transmembrane protein that belongs to the poliovirus receptor-like protein family. It is expressed on T cells and NK cells, but not on monocytes, neutrophils, and B cells. PVRIG binds with high affinity to poliovirus receptor-related 2 (PVRL2/CD112/Nectin-2) and inhibits T-cell mediated immune responses. Expression of PVRIG on T cells is high in multiple cancers such as lung, kidney, breast, ovarian, and endometrial cancers. Blocking of PVRIG alone or in combination with other immune checkpoint proteins such as TIGIT and PD-1 has been demonstrated to increase the production of cytokines and T-cell mediated cytotoxic activity. A novel monoclonal antibody (COM701) has been demonstrated to disrupt the interaction between PVRIG and PVRL2, thus inhibiting tumor growth.
