Description: Interleukin-34 (IL-34) belongs to the short-chain helical hematopoietic cytokine family that promotes the proliferation, survival and differentiation of monocytes and macrophages. It specifically binds CD14+ monocytes, promotes survival and proliferation of human peripheral blood monocytes, and stimulates macrophage colony formation by human bone marrow cells. Clinical studies demonstrated that IL-34 levels significantly increase in the circulation and synovial fluid of patients with rheumatoid arthritis and knee osteoarthritis. Hence it is used as a promising clinical biomarker and therapeutic target for inflammatory arthritis. BioVision’s IL-34 (Human) ELISA Kit is used for the quantitative detection of IL-34 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detectn antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of IL-34 in the sample.
