Description: Matrix metalloproteinase-8 (MMP-8) also known as Neutrophil collagenase or PMNL collagenase belongs to the family of matrix metalloproteinases of zinc- and calcium-dependent endo-peptidases responsible for degradation of extracellular matrix. These are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. MMP-8 is a collagen cleaving enzyme which is present in the connective tissue of most mammals. It can degrade fibrillar type I, II, and III collagens. MMP-8 is an essential mediator in systemic subclinical inflammatory response in obesity, and a potential drug target. Analysis of MMP-8 in breast cancer cell lines revealed a causal connection between MMP-8 activity and IL6 and IL8 production, suggesting a role for MMP-8 in the regulation of the innate immune system. BioVision’s MMP-8 (Human) ELISA Kit is used for the quantitative detection of MMP-8 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of MMP-8 in the sample.
