MUC5B is a heavily glycosylated protein that belongs to the mucin family of proteins. It is present in both upper and lower respiratory tracts. In the upper respiratory tract, it is expressed in oral and nasal glands. In the tracheobronchial conducting airways, MUC5B is expressed in submucosal glands and surface epithelium but is less predominant compared to MUC5AC. In distal airways (such as bronchioles), MUC5B is the predominant mucin. Some of the main functions of MUC5B include mucociliary clearance, regulation of immune responses, and maintaining lung homeostasis. Overexpression of MUC5B may cause reduced mucociliary clearance of inhaled particles, chemicals, and microorganisms from the lungs. This, in turn, may compromise mucosal host defense mechanisms. Increased MUC5B expression may also disrupt repair responses in the alveoli of respiratory bronchioles. Overexpression of MUC5B may contribute to the development of pulmonary diseases such as idiopathic pulmonary fibrosis (IPF), chronic obstructive pulmonary disease (COPD), asthma, and cystic fibrosis (CF). Thus, MUC5B could serve as an efficient target to develop a treatment for pulmonary diseases. BioVision’s MUC5B (Human) ELISA kit is based on the Sandwich ELISA principle to quantitatively measure the concentration of MUC5B in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is proportional to the concentration of MUC5B in the sample or standard.
