The breast cancer metastasis suppressor-1 (BRMS1) is a candidate metastasis-suppressing gene and has been shown to potentially inhibit tumor progression. BRMS1 transcriptional repressor and anoikis regulator protein reduces the metastatic potential, but not the tumorogenicity of human breast cancer and melanoma cell lines. The protein encoded by the BRMS1 gene localizes primarily to the nucleus and is a component of the mSin3a family of histone deacetylase complexes (HDAC). It promotes HDAC1 binding to promoter regions and apoptosis in cells that have inadequate adherence to a substrate, a process called anoikis, and may thereby inhibit metastasis. It also acts as a mediator of metastasis suppression in breast carcinoma. BRMS1 down-regulates expression of anti-apoptotic genes that are controlled by NF-kappa-B. Recombinant human RMS1 protein, fused to T7-tag at N-terminus, was expressed in E. coli.
