Background: Rabbit uterine smooth muscle cells are isolated from uterine Tissue. The uterus is an organ for gestation of the fetus. It is located in the middle of the pelvis, between the bladder and rectum. Its position can vary with the filling degree or position of the bladder and rectum. The normal position of the uterus is mainly maintained by the uterine ligaments, pelvic diaphragm, urogenital diaphragm, and central tendon of the perineum. When these structures are damaged or loose, it can cause uterine prolapse. The myometrium is relatively thick and consists of bundles or slices of smooth muscle. The muscle bundles are separated by connective Tissue. Uterine smooth muscle has contraction function. Contraction is regulated by hormones, and its contraction activity helps sperm to transport to the fallopian tube, discharge through menstrual blood, and deliver the fetus. The smooth muscle layer of the uterus contains a large number of blood vessels. If the smooth muscle layer cells grow excessively, it will inevitably cause the thickening of the blood vessel wall and blockage of the lumen. After 3 days of primary isolation and culture of uterine smooth muscle cells, the cells can be seen to adhere to the flask, and the cell morphology and size are different. Regular, triangular or fan-shaped, with oval and centered nucleus. The cells reach confluence after 2 weeks, most of the cells stretch out in a long fusiform shape, with abundant cytoplasm and branch-like protrusions. The cells are arranged in a single layer or partially overlapped in multiple layers growth, ups and downs. When the cell density is low, they are often intertwined into a network. When the density is high, they are arranged in a whirlpool or fence shape. After passage, the cells grow faster and reach confluence in 4-6 days and maintain the above-mentioned morphological characteristics and growth characteristics.
