GRL-0617 is a potent inhibitor of the papain-like protease (PLpro) from the coronavirus that causes severe acute respiratory syndrome (SARS-CoV). PLpro cleaves the viral polyproteins. GRL-0617 inhibits SARS-CoV and SARS-CoV-2 PLpro in vitro with IC₅₀ values of 0.6 μM and 2.4 μM respectively. GRL-0617 shows antiviral activity with EC₅₀ values of 14.5 μM and 27.6 μM in SARS-CoV and SARS-CoV-2 virus-infected Vero E6 cells.
