Nigericin is a polyether ionophore which disrupts membrane potential and stimulates ATPase activity in mitochondria. Ion selectivity is K⁺> Rb⁺ Cs⁺>> Na⁺. Also activates NLRP3/NALP3 and signals through pannexin-1 to induce caspase-1 maturation and IL-1β processing and release.
