Product Description
The cytokine IL-10 is a key anti-inflammatory mediator with an essential role in the development and proliferation of regulatory T cells, restricting myeloid and chronic inflammatory T cell responses. It has been shown that in solution IL-10 predominantly exists as a homodimer. However, the subunits within the dimer are non-covalently associated, although each subunit contains two intra-chain disulfide bonds. IL10 binds to its hetero-tetrameric receptor comprising IL10RA and IL10RB, which activates multiple pathways that contribute to the downstream anti-inflammatory effects. It also plays important roles in other processes such as sterile wound healing, autoimmunity, cancer, and homeostasis. Given the wide spectrum of the anti-inflammatory properties attributed to IL-10, both IL-10 enhancing and blockade strategies have been tried in a number of diseases and cancer therapy.

Activity (Specifications/test method): The specific activity was determined by the dose-dependent simulation of the proliferation of MC/9 cells (mouse mast cell line) in the presence of IL-4 using BioVisions Quick Cell Proliferation colorimetric Assay Kit Plus (Cat. No. K302). The ED50 is typically in a single digit ~ 1ng/ml.
Biological activity: The ED determined by the ability to induce proliferation of human Interleukin-7 receptor & human TSLP Receptor co-transfected with mouse BaF3 cells was found to be less than 0.3 ng/ml.
Results: <0.3 ng/ml.
