Test Principale: This assay employs a two-site sandwich ELISA to quantitate C-MAPT/C-TAU in samples. An antibody specific for C-MAPT/C-TAU has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyC-MAPT/C-TAU present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for C-MAPT/C-TAU is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of C-MAPT/C-TAU bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview: Tau protein is a highly soluble microtubule-associated protein (MAP) . In humans, these proteins are mostly found in neurons compared to non-neuronal cells. One of tau's main functions is to modulate the stability of axonal microtubules. Tau is not present in dendrites and is active primarily in the distal portions of axons where it provides microtubule stabilization but also flexibility as needed. This contrasts with STOP proteins in the proximal portions of axons which essentially lock down the microtubules and MAP2 that stabilizes microtubules in dendrites. The tau gene locates on chromosome 17q21, containing 16 exons. The major tau protein in the human brain is encoded by 11 exons. Exon 2, 3 and 10 are alternative spliced, allowing six combinations (2-3-10-; 2+3-10-; 2+3+10-; 2-3-10+; 2+3-10+; 2+3+10+) .
Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
