Test Principale: This assay employs a two-site sandwich ELISA to quantitate NKB in samples. An antibody specific for NKB has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyNKB present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NKB is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NKB bound in the initial step. The color development is stopped and the intensity of the color is measured.

Functional studies showed that the mutant protein had 10-fold less activity than wildtype, and the mutation was not found in 100 Turkish controls.It is found in higher concentration in pregnant women suffering pre-eclampsia and can bind the immune-cloaking molecule phosphocholine. Neurokinin B and its receptor, neurokinin 3 receptor, are critical parts of the control system that switches on the master regulator of human puberty, governed by the brain through the release of the hormone GnRH which starts a series of processes that ultimately leads to the production of sex hormones.
Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
