Test Principale: This assay employs a two-site sandwich ELISA to quantitate SPTLC2 in samples. An antibody specific for SPTLC2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anySPTLC2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SPTLC2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SPTLC2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview: Serine palmitoyltransferase (SPT; EC 2.3.1.50) is the key enzyme in sphingolipid biosynthesis. It catalyzes the pyridoxal-5-prime-phosphate-dependent condensation of L-serine and palmitoyl-CoA to 3-oxosphinganine. SPTLC2encodes a long chain base subunit of serine palmitoyltransferase. Serine palmitoyltransferase, which consists of two different subunits, is the initial enzyme in sphingolipid biosynthesis. It catalyzes the pyridoxal 5'-phosphate dependent condensation of L-serine and palmitoyl CoA to 3-oxosphinganine. Mutations in this gene were identified in patients with hereditary sensory neuropathy type I. Alternatively spliced variants encoding different isoforms have been identified.The LCB2 cDNA encodes a protein of 562 amino acids that is more than 90% identical to the mouse protein.
