Description: Matrix metalloproteinase-12 (MMP-12) also known as macrophage metalloelastase or macrophage elastase belongs to the matrix metalloproteinase family. These are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. MMP-12 hydrolysis soluble and insoluble elastin and specific cleavages are also produced at 14-Ala-|-Leu-15 and 16-Tyr-|-Leu-17 in the B chain of insulin. It has significant elastolytic activity and can accept large and small amino acids at the P1' site, but has a preference for leucine. MMP-12 plays an important role in the pathogenesis of several inflammatory diseases such as chronic obstructive pulmonary disease, emphysema, and asthma. MMP-12 acts as a potential biomarker for IS in obese patients. BioVision’s MMP-12 (Human) ELISA Kit is used for the quantitive detection of MMP-12 in serum, plasma, tissue homogenates and other biological fluids. It is based on the principle of sandwich ELISA. The capture antibody is pre-coated on 96-well plates. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin is added and unbound conjugates are washed away with wash buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of MMP-12 in the sample.
