Immunogen: Stimulated human leukocytes were used as the immunogen for the anti-ICAM-3 antibody.
Research Area: Immunology, Signal Transduction
Tested Application: Bl, Flow, IF, IHC-P
Application: Functional Activity: Blocks binding of CD11a (LFA-1) to CD50 (ICAM-3) (Order SAF formulation)
Flow Cytometry: 0.5-1 ug/million cells in 0.1ml
Immunofluorescence: 0.5-1 ug/ml
Immunohistochemistry (FFPE) : 0.5-1 ug/ml for 30 min at RT (1)
Prediluted format: incubate for 30 min at RT (2)
Optimal dilution of the anti-ICAM-3 antibody should be determined by the researcher.
1. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required) , drip mAb solution onto the tissue section and incubate at RT for 30 min.

BACKGROUND: Recognizes the D1 domain of an N-glycosylated glycoprotein of 120kDa with intra-chain disulfide bonds, identified as CD50 or ICAM-3. CD50 is the major ligand for LFA-1 (CD11a/CD18) and may have signaling role to increase adhesion. It is expressed on thymocytes and T lymphocytes and is resistant to treatment with phosphatidylinositol (PI) phospholipase C. This mAb is excellent for staining of formalin/paraffin tissues.
