MDL-800 is a potent and selective allosteric activator (EC50 = 10.3 µM) of SIRT6, a member of the SIRT deacetylase family. Does not display any activities toward SIRT1, SIRT3, SIRT4 and HDAC1-11 at concentrations up to 50 μM or 100 μM. MDL-800 increases the deacetylase activity of SIRT6 by up to 22-fold resulting in a decrease in H3K9Ac and H3K56Ac in hepatocellular carcinoma (HCC) cells, causing cell cycle arrest and inhibiting their proliferation.
