Test Principale: This assay employs a two-site sandwich ELISA to quantitate PIM2 in samples. An antibody specific for PIM2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPIM2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PIM2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PIM2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview: PIM2 encodes a protooncogene that acts as a serine/threonine protein kinase. Studies determined the encoded protein functions to prevent apoptosis and to promote cell survival. The deduced 334-amino acid sequence of the clone showed 90% identity with the mouse Pim2 protein. Like mouse Pim2, the human protein appears to be a serine threonine kinase. Northern blot analysis detected 2 PIM2 transcripts. A 2.2-kb transcript was expressed in all tissues tested, but most abundantly in hematopoietic tissues, spleen, thymus, and peripheral blood leukocytes, as well as in testis, small intestine, and colon. It was also highly expressed in human leukemic and lymphoma cell lines and a colorectal adenocarcinoma cell line. A 5.0-kb transcript was expressed only in spleen, thymus, small intestine, colon, and in very small amounts in peripheral blood leukocytes.
Stability: The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
